The research on several of them is genuinely encouraging. Others are promising but still early. None of them are a substitute for sleep, movement, blood sugar control, or a conversation with your doctor about what's actually driving your symptoms. With that framing in place, let's walk through the evidence for some of the most talked-about options.

* Magnesium L-Threonate: The Form That Actually Reaches the Brain

Not all magnesium is created equal when it comes to brain health. Most forms on shelves, like magnesium citrate or oxide, are absorbed reasonably well in the gut but don't cross the blood-brain barrier in meaningful amounts. Magnesium L-threonate, developed by MIT researchers and often sold under the brand name Magtein, was specifically designed to solve this problem.

The foundational animal research found that this form raised magnesium levels in the brain and increased synaptic density, leading to marked improvements in learning and memory in both young and aged rats. That mechanistic story has since been backed by human trials. A randomized, placebo-controlled study in older adults with signs of cognitive decline found meaningful gains in memory and cognitive function over 12 weeks of supplementation. More recent research has extended this further, with a 2026 randomized trial showing that six weeks of daily supplementation improved working memory, reaction time, and overall cognitive performance, alongside a striking reduction in "cognitive age" as measured against normative data.

It's worth noting that this newer trial was conducted in healthy adults, not people with diagnosed cognitive impairment, and the effect sizes in younger, healthier brains may not generalize perfectly to someone in their sixties with more established decline. Still, the consistency across mechanism, animal data, and multiple human trials makes this one of the more evidence-backed options on this list.

Caveats: Effective doses in the studies were fairly high, typically 1,500 to 2,000 mg of magnesium L-threonate daily (providing a modest amount of elemental magnesium, since threonate itself is a large molecule). Gastrointestinal upset is possible, especially at higher doses. Magnesium can interact with certain antibiotics and osteoporosis medications, so timing matters, and it's worth checking with your prescriber if you take other medications regularly.

* Lion's Mane Mushroom: Encouraging but Still Early

Lion's mane (Hericium erinaceus) has become one of the most popular "brain mushrooms," and the interest isn't baseless. It contains compounds called hericenones and erinacines that stimulate nerve growth factor, a protein involved in the survival and repair of neurons.

The most cited human study is a 16-week trial in adults with mild cognitive impairment, where those taking lion's mane improved more on a standard cognitive scale than the placebo group. A meaningful catch, though, is that those benefits faded within a month of stopping supplementation, suggesting this needs to be taken consistently rather than as a short course. A separate pilot study in people with mild Alzheimer's disease, using an erinacine A-enriched extract over 49 weeks, showed improvement on standard dementia scales as well. Smaller studies in healthy and younger adults have found more modest, mixed effects on cognitive speed, mood, and stress.

Caveats: Trials so far have been small, and long-term safety data is limited. People with mushroom allergies should avoid it. There's some theoretical concern about interaction with blood-thinning medications given effects seen in animal studies, and supplement quality varies widely between brands, so third-party testing matters here more than usual.

* L-Theanine: Calm, Focused Attention, Better Sleep, Less Anxiety
L-theanine, an amino acid found naturally in green and black tea, is one of the more reliable calming compounds in this category. But, before bed, it can help you drift off into dreamland.

Caveats: Generally very well tolerated, though some people notice mild drowsiness. It can modestly lower blood pressure, which is worth knowing if you're already on blood pressure medication.

Human trials specifically testing luteolin for cognition are still sparse. Most of what we know comes from cell and animal studies, plus a handful of small human trials looking at luteolin combined with other compounds for conditions like autism spectrum symptoms and allergic conditions, rather than midlife cognitive complaints directly. It belongs on a watch list of promising compounds rather than a proven intervention yet.

Caveats: Luteolin has poor natural bioavailability, so supplement forms often use special delivery methods to improve absorption (e.g., phytosome or nanoparticle forms); not all products are equivalent. It may interact with medications metabolized through certain liver enzyme pathways, so it's worth mentioning to a pharmacist if you're on multiple prescriptions.

Form matters quite a bit here, and this is where a lot of people get shortchanged by low-quality supplements:

- **B6: The active, coenzyme form is pyridoxal-5-phosphate (P5P). It's more readily used by the body than plain pyridoxine.
- **B12: Methylcobalamin and adenosylcobalamin are the active forms your cells actually use, compared to cyanocobalamin, the cheap synthetic form that requires an extra conversion step your body has to perform. - **Folate: L-methylfolate (5-MTHF) is the active form, and it bypasses the enzyme step that people with common MTHFR gene variants struggle with. This is different from folic acid, the synthetic form added to fortified foods and found in many cheaper multivitamins.

**Caveats: More is not better here. Long-term high-dose B6 (generally above 100 to 200 mg daily for extended periods) has been linked to peripheral nerve damage, sometimes irreversible. Folic acid, especially at high doses, can mask the anemia caused by B12 deficiency while the underlying nerve damage from that deficiency continues untreated, which is one reason checking your actual B12 and homocysteine levels before megadosing is worth doing. **Caveats: Quality and freshness matter a great deal, since oxidized fish oil can do more harm than good. High doses can have a mild blood-thinning effect, which is relevant if you're on anticoagulant medication or scheduled for surgery. And fish oil may stimulate mast cells to release histamine and other inflammatory mediators, if you are sensitive. Algae-based or plant-based flaxseed oil omega-3 supplements are a solid option for anyone avoiding fish.

* Vitamin D: Correcting a Deficiency Matters More Than Chasing High Doses

**Caveats: Vitamin D is fat-soluble and accumulates in the body, so toxicity from excessive supplementation, while uncommon, is possible and can cause elevated calcium levels and related symptoms. Lion's mane and L-theanine have real, if smaller, human trials behind them, with lion's mane needing consistent use to maintain any benefit. Luteolin is a compound worth watching rather than relying on just yet. And omega-3s and vitamin D matter enormously if you're deficient, and much less if you're not, which makes testing before supplementing a genuinely useful step rather than an unnecessary one.
